Mice treated with KCL-286 showed significantly fewer DNA double-strand breaks in their neurons than untreated mice, a type of genetic damage frequently observed in the brains of Alzheimer's patients. The drug also reduced inflammation throughout the brain.
Unlike many experimental Alzheimer's treatments that target a single mechanism, such as amyloid or tau buildup, KCL-286 activates a protein in the retinoic acid pathway, the series of chemical reactions the body uses to process vitamin A, which in turn enhances DNA repair by upregulating a gene called BRCA1.
Because KCL-286 has already cleared an initial Phase 1 human safety trial for its original spinal cord indication, researchers believe it could advance into Alzheimer's clinical testing considerably faster than a novel compound starting from scratch.
